Cetirizine is metabolized to a limited extent by oxidative O-dealkylation to a metabolite with negligible antihistaminic activity. It is the mechanism by which histamine H1-receptor antagonist competes with histamine on effector cells in the gastrointestinal tract, blood vessels, and respiratory tract.
Its contraindication is hypersensitivity to cetirizine hydrochloride or any of its ingredients, levocetirizine, or hydroxyzine, which may cause CNS (central nervous system) depression. Avoid activities requiring mental alertness until accustomed to medication. Cetirizine reduces inflammatory cell recruitment and ICAM-1 (or CD54) expression on conjunctival epithelium in both early and late-phase reactions after allergen-specific challenge.
