Hello doctor,

I checked my CBC both manually as well as through the analyzer. There is a variation in the values of the two by three points. I am confused. The results are as follows:

  1. Analyzer results -16.9.

  2. Manual results -13.4.

My doctor suggested doing JAK 2 tests for further investigation. Kindly help.

Please note that you have not mentioned which parameter you have measured. Is it Hb (hemoglobin) estimation or total WBC (white blood cell) count? Hb estimation is done only through the colorimeter method or by using a cell counter. The total WBC count measured by the cell counter and when checked manually can vary. But, the cell counter reading method is more accurate when compared to the manual method.

Hence, you can rely on cell counter reading if your cell counter is calibrated and if you are maintaining daily quality control of it. Kindly let me know whether you are talking about Hb estimation or WBC count. Also, provide a detailed clinical history of your complaint. JAK (Janus Kinase) 2 mutation study is required for the diagnosis of polycythemia vera.
