My girlfriend had her periods 16 days back but two days back she had a white spotting and today she had blood coming out of her vagina for a second but stopped. Please tell me what does this mean.

The reason for this kind of bleeding can be some abnormal uterine bleeding or simply ovulation bleeding. Today is her 16th day of period probably ovulation day and her detailed obstetric history is not known. So watch for any pain or repeat episode of bleeding and if so get one ultrasound pelvis done.

Does this have any relation to pregnancy because before we had unprotected sex without penetration but she got her periods after that, so is there any chance of pregnancy?

As she got her periods after that so nothing to worry for that episode. But if it was unprotected sex this cycle also then mild bleeding around this day could be a sign of implantation bleeding. Remember even precum can cause pregnancy.

We did have but never rubbed or had penetration and what she had was not a blood flow but just a drop of blood which did not come later on.

It is fine if you did not touch her vagina or any rubbing or penetration. Then you need not worry. Just a drop of blood can be due to any reason. We need to watch until her cycle date is due. If you are too worried about the risk of pregnancy then you can get her blood test beta hCG done one week prior to her next expected date.

Yes, I did rub her vagina with fingers and I am pretty sure that I did not have precum on my fingers and nor did I use my penis to rub. It is that she is having small irritation near her vagina along with vaginal discharge and small bubbles like stuff. What does that mean? It is just that we are anxious.

It is fine. If nothing of the sort, then why to be so anxious and worry about pregnancy? She might be having some vaginal infection (fungal) and nothing else significant. When is her next date due? I do not think that there is anything to worry. But, you still have one more week. So, as I mentioned you can get her blood test for pregnancy one week prior to her expected date and be doubly sure before leaving. And take due precautions now onwards.
